Description PDK1 (3-phosphoinositide-dependent kinase 1), also known as PDPK1, is a key mediator of biological responses downstream of insulin and other tyrosine kinase receptors, regulating cell survival, cell cycle control, protein translation, and glucose metabolism. PDK1, containing a C-terminal pleckstrin homology domain, is activated in a phosphatidyl inositol 3,4,5-trisphosphate-dependent manner and phosphorylates multiple signaling molecules, including PKB/Akt, PKC, p70S6 kinase, serum and glucocorticoid-induced kinase. In T cells, nucleation of TCR-induced signaling complex for NFkappaB activation pathway is another crucial role of PDK1. Subcellular localization of PDK1 depends on conditions and has not been completely understood.
