Description TCF4 (T cell factor 4; also known as TF7L2) serves as transcriptional activator affecting e.g. cell cycle and terminal cell differentiation. In the absence of Wnt signals, TCF4 interacts with TLE/Groucho corepressor, supressing the transcription of TCF4 target genes. Wnt signaling leads to stabilisation and accumulation of beta-catenin, which binds TCF4 N-terminus, whereas TCF4 C-terminal HMG-box domains interact with DNA. Cell death-associated Daxx protein or plakoglobin belong to negative regulators of TCF4-mediated transcription, on the other hand, there is a positive feedback loop of beta-catenin-TCF4 pathway comprising the p300 coactivator.
