| Description | The Thomsen-Friedenreich antigen acts as an oncofetal antigen, with low expression in normal adult tissues but increasing to fetal levels of expression in hyperplasia or malignancy. It is considered as a pan-carcinoma marker. Expression is on the apical surface of mucin-producing normal and cancerous colorectal epithelial cells. Staining is also present in the secreted mucin. Mouse Monoclonal Antibody to Thomsen-Friedenreich Pan-Tumor Antigen / CD176. The antigen / epitope is mainly found on the cell membrane. A78-G/A7 binds to CD176 cell surface antigen / asialoglycophorin but not to glycophorin. It agglutinates neuraminidase- treated but not untreated human erythrocytes. The epitope is sensitive to periodate oxidation. |

| Specificity | Positive reactions are found for primary breast cancer and colorectal cancer and metastases derived therefrom. Normal colonic adjacent tissue is negative, whereas breast epithelium may react, esp. luminal membranes. In immunocytochemistry positive staining is also found with acute myelogeneous leukemia cells (HL-60, KG-1). In immunoblotting a ma- jor band of 155 kDa is detectable. Specific binding with both anomers of TF as well as with the TFÌÄü-related asialo-GM1. |
