| Description | Chicken Endoribonuclease Dicer (DICER1) ELISA Kit has high sensitivity and excellent specificity for detection of Chicken DICER1. No significant cross-reactivity or interference between Chicken DICER1 and analogues was observed. Endoribonuclease Dicer (DICER1) is a double-stranded RNA (dsRNA) endoribonuclease playing a central role in short dsRNA-mediated post-transcriptional gene silencing. Cleaves naturally occurring long dsRNAs and short hairpin pre-microRNAs (miRNA) into fragments of twenty-one to twenty-three nucleotides with 3' overhang of two nucleotides, producing respectively short interfering RNAs (siRNA) and mature microRNAs. SiRNAs and miRNAs serve as guide to direct the RNA-induced silencing complex (RISC) to complementary RNAs to degrade them or prevent their translation. Gene silencing mediated by siRNAs, also called RNA interference, controls the elimination of transcripts from mobile and repetitive DNA elements of the genome but also the degradation of exogenous RNA of viral origin for instance. The miRNA pathway on the other side is a mean to specifically regulate the expression of target genes. This Chicken Endoribonuclease Dicer (DICER1) ELISA Kit employs a two-site sandwich ELISA to quantitate DICER1 in samples. An antibody specific for DICER1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyDICER1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for DICER1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of DICER1 bound in the initial step. The color development is stopped and the intensity of the color is measured. |
