| Description | Bovine Maspardin (SPG21) ELISA Kit has high sensitivity and excellent specificity for detection of Bovine SPG21. No significant cross-reactivity or interference between Bovine SPG21 and analogues was observed. Maspardin (SPG21) binds to the hydrophobic C-terminal amino acids of CD4 which are involved in repression of T cell activation. The interaction with CD4 is mediated by the noncatalytic alpha/beta hydrolase fold domain of Maspardin (SPG21). It is thus proposed that Maspardin (SPG21) product modulates the stimulatory activity of CD4. Mutations in this Maspardin (SPG21) are associated with autosomal recessive spastic paraplegia 21 (SPG21), also known as mast syndrome. Alternative splicing results in multiple transcript variants. This Bovine Maspardin (SPG21) ELISA Kit employs a two-site sandwich ELISA to quantitate SPG21 in samples. An antibody specific for SPG21 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anySPG21 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for SPG21 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of SPG21 bound in the initial step. The color development is stopped and the intensity of the color is measured. |
