| Description | Bovine Mucin-1 (MUC1) ELISA Kit has high sensitivity and excellent specificity for detection of Bovine MUC1. No significant cross-reactivity or interference between Bovine MUC1 and analogues was observed. Mucin-1 (MUC1) encodes a membrane-bound protein that is a member of the mucin family. Mucins are O-glycosylated proteins that play an essential role in forming protective mucous barriers on epithelial surfaces. These proteins also play a role in intracellular signaling. MUC1 is expressed on the apical surface of epithelial cells that line the mucosal surfaces of many different tissues including lung, breast stomach and pancreas. MUC1 is proteolytically cleaved into alpha and beta subunits that form a heterodimeric complex. The N-terminal alpha subunit functions in cell-adhesion and the C-terminal beta subunit is involved in cell signaling. Overexpression, aberrant intracellular localization, and changes in glycosylation of this protein have been associated with carcinomas. MUC1 is known to contain a highly polymorphic variable number tandem repeats (VNTR) domain. Alternate splicing results in multiple transcript variants. This Bovine Mucin-1 (MUC1) ELISA Kit employs a two-site sandwich ELISA to quantitate MUC1 in samples. An antibody specific for MUC1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyMUC1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MUC1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MUC1 bound in the initial step. The color development is stopped and the intensity of the color is measured. |
