Description Endothelin-1 (ET-1), a peptide of 21 amino acid residues, is the most potent vasoconstrictor substance known. The discovery by Rubanyi's group in 1985 of a peptide termed endothelium-derived contracting factor (EDCF) and the isolation, sequencing, cloning and naming endothelin by Yanagisawa of the most potent vasoconstrictor has led to more understanding of many physiological effects. ET-1 is known to be one of a family of three mammalian vasoactive peptides including endothelin-2 (ET-2) and endothelin-3 (ET-3). These related peptides differ from ET-1 at the two and six amino acid residue positions, respectively. Those genes make three pre-propeptides that are cleaved into three big endothelin molecules, each 39 amino acid long. A number of reviews of function, molecular biology of the endothelin family of peptides and propeptides are available. ET-1 has been shown to have potent effects on smooth muscle cells, fibroblasts and to be involved in many disease processes, particularly cardiovascular diseases. It has been shown to be important in congestive heart failure, renal failure and pulmonary hypertension. Human ET-1 has identical amino acid sequence to bovine, canine, murine, porcine, rabbit and rat ET-1. This kit is a solid phase sandwich ELISA using 2 kinds of high specific antibodies. Tetra Methyl Benzidine (TMB) is used as coloring agent (Chromogen). The strength of coloring is in proportion to the quantities of ET-1.
