Description Bovine Acetyl-CoA Carboxylase Synthetase (ACC) ELISA Kit has high sensitivity and excellent specificity for detection of Bovine ACC. No significant cross-reactivity or interference between Bovine ACC and analogues was observed. Acetyl-CoA carboxylase (ACC) is a biotin-dependent enzyme that catalyzes the irreversible carboxylation of acetyl-CoA to produce malonyl-CoA through its two catalytic activities, biotin carboxylase (BC) and carboxyltransferase (CT). ACC is a multi-subunit enzyme in most prokaryotes and in the chloroplasts of most plants and algae, whereas it is a large, multi-domain enzyme in the endoplasmic reticulum of most eukaryotes. The most important function of ACC is to provide the malonyl-CoA substrate for the biosynthesis of fatty acids. The activity of ACC can be controlled at the transcriptional level as well as by small molecule modulators and covalent modification. The human genome contains the genes for two different ACCs — ACACAand ACACB. This Bovine Acetyl-CoA Carboxylase Synthetase (ACC) ELISA Kit employs a two-site sandwich ELISA to quantitate ACC in samples. An antibody specific for ACC has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyACC present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for ACC is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of ACC bound in the initial step. The color development is stopped and the intensity of the color is measured.
