| Description | Bovine Heat shock protein beta-1 (HSPB1) ELISA Kit has high sensitivity and excellent specificity for detection of Bovine HSPB1. No significant cross-reactivity or interference between Bovine HSPB1 and analogues was observed. Heat shock protein (HSP) 27 is one of the small HSPs. Like other small heat shock proteins, HSP27 is regulated at both the transcriptional and posttranslational levels . In response to stress, the expression level of HSP27 increases several-fold to confer cellular resistance to the adverse environmental change. HSP27 is phosphorylated at Ser15, Ser78 and Ser82 by MAPKAP kinase 2 as a result of the activation of the p38 MAP kinase pathway . Phosphorylation of HSP27 causes a change in its tertiary structure, which shifts from large homotypic multimers to dimers and monomers . It has been shown that phosphorylation and increased concentration of HSP27 modulates actin polymerization and reorganization . This Bovine Heat shock protein beta-1 (HSPB1) ELISA Kit employs a two-site sandwich ELISA to quantitate HSPB1 in samples. An antibody specific for HSPB1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyHSPB1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for HSPB1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of HSPB1 bound in the initial step. The color development is stopped and the intensity of the color is measured. |
