| Description | EliKine™ Human IL-4 ELISA Kit has high sensitivity and excellent specificity for detection of Human IL-4. No significant cross-reactivity or interference between Human IL-4 and analogues was observed. IL-4 is a 129 amino acids long secreted protein. IL-4 is a cytokine produced mostly by activated T lymphocytes, mast cells and basophils. It has variety of immune response modulating functions by acting on number of cell types. IL-4 participates in several B-cell activation processes. It is a co-stimulator of DNA-synthesis. It induces the expression of class II MHC molecules on resting B-cells. It enhances both secretion and cell surface expression of IgE and IgG1. It also regulates the expression of the low affinity Fc receptor for IgE (CD23) on both lymphocytes and monocytes. IL-4 positively regulates IL31RA expression in macrophages. EliKine™ Human IL-4 ELISA Kit employs a two-site sandwich ELISA to quantitate IL-4 in samples. An antibody specific for IL-4 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any IL-4 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for IL-4 is added to the wells. After washing, proprietary EliKine™ Streptavidin-HRP conjugates is added to the wells. Following a wash to remove any unbound streptavidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of IL-4 bound in the initial step. The color development is stopped and the intensity of the color is measured. |
